My wife is starting a new position at Target as a team lead (not ETL though)...wondering when she will be elgibile for vacations and paid holidays?

If she works over 30 hours a week yes. She will get two weeks of vacation and 3 personal days

Does she have to wait for the 6 months to begin accruing vacation or does the accrual start on day 1? (Or are they days already accrued on day 1?)...just trying to plan out when we'll be able to take vacation together again lol.

Those are your fun months 🙂 lol. For the most part around that time all vac is blacklisted (even for us ETL's).
